ABSTRACT
Background: Migraine has recently become a major interest to neuroscientists. Almotriptan is an effective medicine used to treat migraine and certain other headaches. This study for the first time has been done to prepare and evaluate specific Almotriptan nanoparticles (A-NP) by different technique. Almotriptan nanoparticles has been developed by different method for antimigraine activity. Polymers such as Chitosan, Albumin, Gelatin, Eudragit-L100 and cross linking agent, were used at different levels to prepare Almotriptan nanoparticles with high entrapment efficiency and low particle size. Completion of the reaction was confirmed by Infrared spectroscopy. Nanoparticles prepared by different methods were evaluated for shape, surface morphology, Polydispersity index, Zeta potential, particle size distribution and entrapment efficiency. Results: Among all the formulation, (AC5) formulation of Almotriptan-loaded Chitosan nanoparticles using ion gelation technique has small particle size around 75.3 nm, Polydispersity index of 0.54, Zeta potential +25.3 mV and high entrapment efficiency around 75.4 %. So it can be the appropriate method for preparing Almotriptan nanoparticles. Conclusion: Chitosan Nanoparticles in the presence of 0.75 % (W/V) Sodium Tripoly-Phosphate as a cross linking agent gives better result and could be used as drug delivery vehicle.
