ANTIOXIDANT EFFECT OF CLERODENDRUM SPLENDENS IN TESTICULAR MILLIEU IN MALE ALBINO RATS

ABSTRACT
This study investigated the potency effect of Clerodendrum splendens on the testes of rats. Twenty one (21) male rats of average weight between 180 to 200g were distributed into seven groups of three rats per group. Rats in group 1 (control group) were administered diet and water only, while groups 2 to 7 were orally administered varying dosage (10, 100, 1000, 1600, 2900 and 5000)mg/kg body weight once daily for 14 days. On the 24th day, the animals were sacrificed by cervical dislocation. Testicular antioxidant level was determined by measuring Superoxide dismutase, reduced glutathione and glutathione-S-transferase activity. The results revealed that the testicular activities
of the enzymic antioxidants GST, SOD and GSH were significantly decreased in rats administered various doses except at 1000mg/g body weight of the extract of clerodendron splendens. GSH activity decreased with 2.12mg/g, 10.42mg/g, 11.06mg/g, 8.51mg/g, 6.38mg/g in the groups administered with different doses of (10, 100, 1600, 2900, 5000) mg/kg body weight. The activity of testicular SOD also decrease by 8.93μmol/min/g, 43.78μmol/min/g, 46.46μmol/min/g,35.74μmol/min/g, 26.80μmol/min/g in the groups administered with different doses of (10, 100, 1600, 2900, 5000) mg/kg body weight. Testicular GST activity was significantly reduced by 0.44μmol/min/g, 1.90μmol/min/g, 2.43μmol/min/g, 1.50μmol/min/g, 1.10μmol/min/g in the groups administered with different doses of (10, 100, 1600, 2900, 5000) mg/kg body weight respectively. The study concluded that the ethanolic extract and fractions of Clerodendrum splendens had deleterious effects on the testes and accessory organs of the animals.
