TOTAL ANTIOXIDANT ACTIVITY OF G?q-RGS2 LOOP ACTIVATOR-AN IN-VITRO STUDY

ABSTRACT
The aim of study was to evaluate the total antioxidant activity of pure chemical moiety (Gαq-RGS2 loop activator) against the DPPH, ABTS, FRAP, iron chelating, superoxide scavenging and nitrite scavenging assay. Trolox was selected as standard antioxidant. The stock solution of trolox and Gαq-RGS2 loop activator 1 mM was prepared in absolute DMSO and working solution of trolox and Gαq-RGS2 loop activator (10, 25, 50 and 100 μM) were made from the stock solution by dilution with required quantity of double distilled water. In results, Gαq-RGS2 loop activator and trolox showed dose dependent (10, 25, 50, 100 μM) anti-oxidant activity in the DPPH, ABTS, FRAP, superoxide
scavenging assay and nitrite scavenging assay except only iron chelating assay. Trolox showed the dose dependent anti-oxidant activity in iron chelating assay but Gαq-RGS2 loop activator showed only at high concentration in iron chelating assay. In conclusion, Gαq-RGS2 loop activator (100 μM) showed an anti-oxidant activity that is similar to trolox ~100 μM for scavenging organic free radical, superoxide radical, cationic radical, peroxyl radical, ferric reduction and chelation capacity. This anti-oxidant activity may be due to electron, nitric oxide donating and free radical capturing in lipophilic-hydrophilic environment and iron chelation properties.
