EFFECT OF CREMOPHOR RH 40 ON DISSOLUTION RATE OF ATORVASTATIN TABLET

ABSTRACT
Atorvastatin is one of the active substance with anti hypercholestrol activity in the statin group. Atorvastatin itself and its active metabolite that is structurally similar with HMG-CoA compete to bind to the active site of HMG-CoA reductase. In the intestine, Atorvastatin undergoes eflux caused by P-glycoprotein and is then taken out of the cell. While Cremophor RH 40 is a substrate of P-Glycoprotein then added into atorvastatin formula. When testing of atorvastatin tablets was performed using the dissolution tool type II on minutes 30 in buffer pH 6.8 it was found that the tablet atorvastatin calcium with 1.5% Cremophor RH 40 had a dissolution of 87.34% whereas without the addition Cremophor RH 40, they were dissolved by 60.2%.
