QUICK IN-VITRO MULTIPLE SHOOT PROLIFERATION AND PLANT REGENERATION FROM THE SEEDS OF MURRAYA KOENIGII (L.) SPERNG

ABSTRACT
An efficient in-vitro propagation protocol for plant regeneration from seeds of Murraya Koenigii was developed by culturing on Murashige and Skoog`s (MS) medium supplemented with Picloram (0.2mg/l, 0.5mg/l), TDZ (0.2mg/l, 0.5mg/l), Zn (0.2mg/l, 0.5mg/l), BAP (0.5mg/l), IBA (0.5mg/l, 1.0mg/l) and 2.0% sucrose. The frequency of shoots regenerated was higher on similar medium in subsequent subcultures. The elongated shoots were then rooted on half strength MS medium supplemented with NAA (2.0mg/l).The complete plantlets were hardened and acclimatized in plant growth chamber at 26±2 ̊C temperature, 80% humidity. Finally the plantlets were transferred in the field condition, where they were survived with 80% survival rate. Growth of plantlet regenerated from seeds in Cocopeat and Soil was also observed.
