STUDY OF GLUCOSE UPTAKE ACTIVITY OF AEGLE MARMELOS ETHANOLIC EXTRACT ON LIVER CELL LINE BY FLOW CYTOMETRY

ABSTRACT
In the present study, we have identified and functionally characterized crude ethanolic leaf extract which increases the glucose uptake in liver cell line HepG2. 2-NBDG, (2-(N-(7-Nitrobenz-2-oxa-1,3-diazol-4-yl)Amino)-2-Deoxyglucose)), a fluorescent deoxyglucose analog, was used as a probe for the detection of glucose taken up by cultured cells. It is transported by both SGLTs and GLUTs and has allowed the study of the effects of drugs targeting glucose uptake and glycolysis. NBDG fluorescence typically displays excitation/emission maxima of ~465/540 nm and can be visualized using optical filters designed for fluorescein. Ethanolic extract of Aegle marmelos increased the NBDG
uptake in HepG2 cells significantly as compared to the control and Metformin treated cells.
