ANALYTICAL METHOD DEVELOPMENT AND VALIDATION FOR SIMULTANEOUS ESTIMATION OF CATECHIN AND CURCUMIN BY HPLC IN AN AYURVEDIC FORMULATION

ABSTRACT A simple, selective, particular and reproducible Reverse phase HPLC technique has been developed for the simultaneous estimation of Catechin and Curcumin from ayurvedic formulation. The method was validated using parameters such as linearity, specificity, precision, accuracy, limit of quantification, limit of detection and robustness as per ICH guidelines. The method was carried out on a Hemochrom Intsil [(C18 (250 X 4.6 mm, 5 μm)] column with a mobile phase consisting of methanol: 0.1% orthophosphoric acid at a flow rate of 0.8 ml/min. Detection was carried out at 269 nm. The retention time of Catechin and Curcumin was 3.169 and 7.669 min, respectively. The regression analysis data indicated good linear relationship for the calibration plots for Catechin and Curcumin in the range of 50-110μg/ml and 2-14 μg/ml and regression coefficient was 0.993 and 0.992 respectively. The proposed method can be used for the estimation of these markers in combined ayurvedic formulation.
