A REVERSED-PHASE HIGH PERFORMANCE LIQUID CHROMATOGRAPHIC METHOD FOR DETERMINATION OF CHLORTHALIDONE IN BULK AND PHARMACEUTICAL DOSAGE FORM

ABSTRACT
A simple, specific, sensitive, precise, and accurate high performance liquid chromatography method was developed for the determination of Chlorthalidone in pharmaceutical tablet dosage form. Chlorthalidone is a diuretic drug frequently prescribed by Indian physicians for treatment of cardiac, hepatic, renal and pulmonary diseases. The method was carried out on reverse phase Prontosil C-8 column (Agilent Infinity) (250×4.6mm, 5μm particle size) using a mixture of methanol: acetonitrile :water in the ratio of (60:20:20 v/v/v ) as mobile phase. The detection was carried out by UV-detector at 275 nm at column temperature 25±2°C. The calibration curve was found to be linear in the range of 2 to 18 μg/ml, in the linearity study regression equation was found to be y = 27694x – 36286 and correlation coefficient was found to be 0.999. Accuracy study was done in 3 different concentration level i.e. 80%, 100%, 120% and % recovery of the method was found to be 100.03%, 99.968% and 100.08% respectively in 3 different levels. This method was Rugged and Robust in different testing criteria, LOD and LOQ were found to be 0.000001898 and 0.000005744 respectively.
