EFFECTS OF HYDRO-ALCOHOLIC EXTRACT OF MEDICAGO SATIVA ON THE CHEMICAL BURN INJURIES IN RATS

ABSTRACT The aim of the present study was to investigate the efficacy of Medicago sativa (MS) extract after topical application on superficial partial thickness burn wound model in rat. Materials and Methods: Fresh plant material was collected from Dehdasht, Iran and extracted with 70% (v/v) ethanol, using maceration method. Twenty-four adult male Wistar rats were randomly divided into 4 groups. Burn wound was created by NaOH upon the shaved skin. Rats in Group-I were treated with simple base cream as a negative control, Group-II was treated by the standard ointment drug (1%Silver Sulfadiazine), Groups-III and IV were treated with simple ointment base containing of 7.5mg and 15 mg % extract which applied to the burns by a sterile applicator. A 10% homogenate of granulation rat tissue was prepared on 11Th day experiment.Reduced glutathione (GSH), malondialdehyde (MDA) and nitric oxide (NO) were determined. All data was expressed as mean ± SD, and statistical significance between experimental and control values was analyzed by ANOVA. Results: Ms at both low and high doses of 7.5 and 15 mg% in creame base significantly increased the level of GSH in burn wound in rat skin tissue compared to the standard drug. A significant decrease of MDA in skin tissues was reported in treatment groups both low and high doses compare to control and standard groups (P<0.01). The concentration of NO was noticeably decreased in the skin homogenate of rats treated with both low and high doses MS extracts and standard groups when compare to control group (P<0.01). Conclusion: The decrease in MDA and NO and increase in GSH levels in low and high MS extract doses reveals wound healing may be due to its antioxidant property.
