BIOISOTERISM REVIEW- AN BIOLOGICAL MODIFICATION

ABSTRACT
The concept of bioisosterism has helped considerably over the years in the search for and design of new drugs. It is a concept, which has been useful in the absence on more precise knowledge of how to failor a chemical structure to produce and required to biological effect. The discoverer of a compound with promising medicinal activity leads to a search for other structurally closely related compounds, which have improved therapeutic properties and reduced unwanted side effects. In the past, medicinal chemists have developed a considerable amount of intuition, in this respect, in selecting appropriate structural modifications. Much of the search has been based on bioisosteric relations and has led to fruitful yield of new, improved drugs. Bioisosterism represents one approach used by the medicinal chemists for the rational modification of lead compounds into safer and more clinically effective agents. The concept of bioisosterism is often considered to be qualitative and intuitive.
