Abstract

EFFECT OF PLANT HORMONE GIBBERELLIC ACID-3 ON RAT TESTICULAR BIOMARKERS AND ITS HISTOLOGY

ABSTRACT

Gibberellic acid-3(GA3) is a Plant Growth Regulator (PGR) used in agriculture. It is a potent metabolic constituent of plant cells and hence of plant foods and herbal medicines. Recent research has indicated that PGRs have potential to regulate animal cell function. Present study was therefore aimed at exploring the short term (acute) effect of GA3 on testicular biochemistry and its histology employing adult male albino wistar rat (160-180 grams) administered varying doses of GA3 subcutaneously. Testes from experimental rat were removed 4 hr following use of anaesthetic ether and were assessed for changes in specific marker enzyme activity. The activities of aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP), gamma glutamyl transpeptidase (GGT) & Amylase yielded differential response to the GA3 doses used. Levels of Cholesterol, Triglyceride, Protein, Albumin, Urea, Uric acid and Creatinine were augmented with that of malondialdehyde (MDA) and glutathione peroxidise (GPx) at all doses of GA3 used. In contrast the activities of superoxide dismutase (SOD) and Catalase (CAT) and reduced glutathione (GSH) level reduced significantly. Deoxyribonucleic acid (DNA) content decreased significantly but no change in level of ribonucleic acid (RNA) was noted. Results indicated that GA3 induced oxidative damage in rat testicular cells and augmented transamination, dephosphorylation and amylolytic activities. GA3 decreased the cellular antioxidant defence potential as also the DNA content. Spectrums of histological aberrations were noted due to the GA3 effect. GA3 caused testicular toxicity through peroxidative damage and raised cellular metabolic activity. GA3 suppressed testicular antioxidant catalytic potential and promoted DNA damage.
