Abstract

EFFECT OF POLYMERS ON CRYSTALLO-CO-AGGLOMERATES OF FENOFIBRATE

ABSTRACT

The crystallo-co-agglomeration is novel particle engineering technique which produces agglomerates with improved solubility, flow properties and compression properties. The purpose of this research work was to study the effect of polymers on Crystallo-Co-agglomerates of fenofibrate, a BCS class II drug with poor compression and dissolution properties. The agglomerates were prepared in a three solvent system where in acetone was used as a good solvent, water as bad solvent and DCM as bridging liquid. The polymers used were Polyvinyl Pyrrolidone (PVP), Polyethylene glycol (PEG6000) and Hydroxypropyl methyl cellulose (HPMC). The flow (Carr index), compression (MyP) and dissolution (% release after 2 hrs) for fenofibrate (40.81,5.9503,8.72%), PVP agglomerates (11.11%,2.386,43.6%)for PEG agglomerate (13.33%,3.5768,60.8%) and HPMC agglomerate (11.11%,3.4121,50.3%).This shows that PEG agglomerate is better in all respect over other two. Thus PEG agglomerates were found moresuitable for preparation of agglomerates of fenofibrate using CCA technique.
