DESIGN, FORMULATION AND IN - VITRO EVALUATION OF OLANZAPINE MATRIX SUSTAINED RELEASE TABLETS USING NATURAL POLYMERS

ABSTRACT
Olanzapine is a thiaenobenzodiazepine derivative belongs to the class
of second generation atypical anti-psychotic agents used for the
treatment of schizophrenia, depressive episodes associated with bipolar
disorder. The present study was to formulate and in-vitro evaluation of
Olanzapine using natural polymers like Sodium alginate, Guar gum,
Aegle Marmelos(Bael fruit) in the ratio of 1:2.5, 1:5, 1:7.5 by Direct
Compression method. Each formulation was evaluated for various pre
and post compression parameters such as flow property, Bulk density,
Tapped density, Weight variation, Swelling index, Hardness, Friability,
Assay, in-vitro dissolution. Among the 9 formulations, F8 formulation
with 1:5 ratio of Olanzapine with Aegle marmelos showed the best
dissolution rate studies. In-Vitro dissolution studies showed (99.93%) sustained release of
drug up to 12 hours and the mechanism of drug release from the Sustained Release tablets
was followed to be zero order kinetics with non-fickian diffusion. FTIR studies showed there
was no interaction between the drug and polymers (or) excipients.
