COMPARISON THE CYTOTOXICITY OF AFLATOXIN B1 AND ZEARALENONE IN L20 B CELLS

ABSTRACT Aflatoxin B1and Zearalenone are important food-borne mycotoxins causing a risk factor from both public health and agricultural perspectives. In this in vitro study, we compared the cytotoxic effect of Aflatoxin B1 with zearalenone on L20 B cell line. No concentrations of Aflatoxin B1 showed a significant reduction of the cellular viability. There were some cytological changes. At low concentration, large numbers of cells were retaining their normal features and there were a few dead cells and pyknosis with cellular debris. The cellular growth inhibition and earlier stages of cytolysis were noticed at a medium and high concentrations (0.156 - 1.25) ng/ml. An increase in cell proliferation was observing in the presence of Zearalenone at (2.5 ng/ml - 0.625 ng/ml) concentrations. The induction rates were: (139.64- 200) % respectively. The stimulated cells were monolayers with overlapping of small dark proliferating cells and abnormal mitotic figures comparison with complete confluent monolayer of large cohesive control group cells. While the lower concentrations of Zearalenone (0.156 ng/ml -0.039 ng/ml) reduced cell growth. The cytologic changes were: high nucleus to cytoplasm ratio, irregular, hyperchromatic nuclei with some dead cells. In conclusion, higher concentrations of Zearalenone were more cytotoxic effects on cells compared with the same concentrations of Aflatoxin B1.
