Abstract

FORMULATION AND EVALUATION OF GLIMEPIRIDE MATRIX TABLETS WITH VARIOUS COMBINATIONS OF POLYMERS.

ABSTRACT

The main aim of the present work was prepare sustained release matrix tablets of glimepiride by using various concentration of polymer. Glimepiride is the first III generation sulphonyl urea it is a potent sulphonyl urea with long duration of action. Glimepiride is poorly water soluble drug, so solubility is the main constraint for its oral bioavability. Glimepiride exhibits very poor solubility at 370C (< 0.004 mg/ml). In media pH > 7 solubility of drug is slightly increased. The rational of this study was to enhance the solubility of the drug by adding β-CD and HP- β-CD. The glimepiride tablets were prepared by direct compression method. Eight formulations were developed with varying concentration of polymers like PVA, HPMC K4, HPMC K100 and Carbopol 934. The tablets were subjected to Pre-compression and post-compression studies, in-vitro drug release and kinetic studies. FTIR and DSC studies shown there was no interaction between drug and polymers. Pre-compression and post-compression properties of tablets were found within limit. A result of the present study indicates the suitability of the above mentioned polymers in the preparation of glimepiride tablets. All the data (K) were found to be significant at the level of p value < 0.005 assessed by two tails ANOVA.
