NEPHROPROTECTIVE EFFECT OF ETHANOLIC EXTRACT OF FLOWERS OF MICHELIA CHAMPACA AGAINST CISPLATIN-INDUCED

ABSTRACT
The present study was designed to evaluate the Nephroprotective effect
of Michelia Champacaflowers ethanolic extract in Cisplatin-induced
nephrotoxicity. Wistar rats were divided randomly into four groups of
six animals each (n=6) receiving different treatments, consisting of
vehicle (normal saline 2ml/kg b.w), single dose of cisplatin (5mg/kg of
body weight; i.p), standard polyherbal drug cystone at a dose of 5ml/kg
body weight,Michelia Champaca flowers ethanolic extract (250mg/kg
body weight) for 14 days. Nephroprotective activity was assessed by
estimating various biochemical parameters likeserumurea, creatinine,
Blood urea nitrogen (BUN), uric acid, total protein and haematological
estimations like Hemoglobin (Hb), packed cell volume (PCV), mean
corpuscular volume (MCV), mean corpuscular hemoglobin
concentration (MCHC), Total leukocyte count (TLC) and Red blood cells count
(RBC).Histological analysis of rat kidneys and body weights were also observed.
Measurement of antioxidant parameters- lipid peroxidation, catalase, Superoxide dismutase
and reduced glutathione was also investigated. Administration of Michelia Champacaflowers
ethanolic extract produced significant nephroprotective activity in cisplatin induced
nephrotoxicity model as evident by decrease in elevated serum creatinine, urea, BUN, uric
acid and total protein levels, which was further confirmed by histopathological study. The
nephroprotective activity of Michelia Champacacould be due to the inherent antioxidant and
free-radical-scavenging principle(s) contained in the extract.
