CLONING, SEQUENCING AND BIOINFORMATIC ANALYSIS OF P1 GENE OF BANANA BRACT MOSAIC VIRUS (BBRMV) ISOLATES

ABSTRACT
Banana bract mosaic disease caused by banana bract mosaic virus is one of the serious viral diseases of banana in India. Bract mosaic infected banana leaf samples were collected from different geographical regions. The sample was initially tested by Direct Antigen Coating Enzyme Linked Immunosorbent Assay (DAC-ELISA) using recombinant polyclonal antiserum to detect BBrMV. PI genes of BBrMV were amplified. The amplified gene was cloned into a T tailed based vector. The clones were sequenced the sequence length of the PI gene of BBrMV was 987 nucleotides (nt) and encoding a protein of 329 amino acids (aa) having a deduced MW of 36kDa was amplified from BBrMV infected banana sample. The genetic diversity and phylogenetic analysis revealed that P1 gene of
TRY isolate of Trichy 93% at nucleotide level (nt) and 88.0% at amino acid (aa) level. When Trichy isolate compared with the other potyvirus group sequences, the nt and aa identity was ranged 16.0-38.0% and 6-15.0% respectively
