SIMULTANEOUS DETERMINATION OF LAMIVUDINE, ZIDOVUDINE AND NEVIRAPINE BY USING REVERSE-PHASE HIGH PERFORMANCE LIQUID CHROMATOGRAPHY

ABSTRACT
A simple, rapid, precise and accurate reverse phase-liquid-chromatographic.[1] method was developed and validated for the Simultaneous Determination of Lamivudine, Zidovudine and Nevirapine.[2] An ODS C18(4.6 x 150mm, 3.5m) kept at ambient temperature, using a mobile phase consisting of Ammonium acetate buffer: Aceto nitrile:Methanol (50:30:20), of pH 5.5, at a flow rate of 1.2 ml/min and UV detection 270nm for Lamivudine, 265 nm for Zidovudine, 230 nm for Nevirapine and 260nm for the combination. The retention time was found to be 2.4min for Lamivudine, 3.3 min for Zidovudine, 4.8 min for Nevirapine. Validation method was carried out as per ICH guidelines. The correlation coefficient for linear curve obtained between concentrations Vs area for standard preparations of
Lamivudine, Zidovudine and Nevirapine is 0.9997, 0.9998 and 0.9985 respectively. Due to its simplicity, rapidness and high precision, the method was successfully applied to the Simultaneous Determination of Lamivudine, Zidovudine and Nevirapine.
