Abstract

PHYTOCHEMICAL EVALUATION AND FINGERPRINT PROFILING OF TECTONA GRANDIS LINN. BY HPTLC AND HPLC

ABSTRACT

Tectona grandis Linn. commonly known as Teak or sagwan considered as a major constituent in many folklore medicines. Leaves of tectona grandis Linn. are mainly used for haemostatic, depurative, anti-inflammatory and vulnerary. They are also useful in inflammations, leprosy, skin diseases,pruritus, stomatitis, indolent ulcers, haemorrhages and haemoptysis.[1,2] Phytochemical screening and chromatographic fingerprint analysis represents a comprehensive qualitative approach for the purpose of species authentication, evaluation of quality and ensuring the consistency and stability of herbs, herbal drugs and their related products. Phytochemical screening gives the idea about basic types of phytochemical presence in plants. An attempt has been made for phytochemical screening of tectona grandis Linn. leaves powder followed by its HPTLC and HPLC fingerprinting. For HPTLC, acidified methanol extract of tectona grandis Linn. leaves powder was subjected to double development on HPTLC plate. First development was carried out using PET ether (60-80) :Ethyl acetate : Formic acid (7:3:0.5) v/v up to 50 mm development, saturation time – 15 minutes with Whatman filter paper 1, then second development was carried out by using PET ether (60-80) :Ethyl acetate : Formic acid (8.5:1.5:0.5) v/v, and then it scanned under UV at 254 nm, 366nm and under visible light (580nm). For HPLC, mobile phase A: 0.1% formic acid in water and B: Methanol were used and separation was performed on C18 column using gradient program. Hence, it was concluded that the phytochemical screening along with established HPTLC and HPLC fingerprints provide technical support for routine quality control, species identification, authentification and is appropriate for standardization
