Abstract

SUBACUTE INTRAVENOUS TOXICITY STUDY OF DISODIUM EDTA IN SWISS ALBINO MICE

ABSTRACT

The study was conducted to determine the sub acute toxicity of disodium EDTA upon slow intravenous injection in mice. Animals were divided into six groups and doses were administered for a period of 14 days in all the groups by slow intravenous injection. G1 was the vehicle control group. G2, G3 and G4 were the disodium EDTA treated (15, 30 & 60 mg/kg) group respectively. Groups G5 and G6 were the recovery groups observed for an additional period of 14 days after discontinuation of dosing. Clinical signs were recorded once daily in all the animals until their scheduled necropsy. Biochemical and hematological evaluations were performed on day 7 and 14 for animals of G1-G4 group and on day 28 for animals of G5 and G6 group. Gross pathology was performed and organs were collected for histopathology on terminal day. Results of the study suggested that there were no visible signs of toxicity in any group treated with disodium EDTA. Biochemical and hematological parameters were comparable to vehicle control groups in the main and recovery groups. Interim or terminal sacrifices did not reveal any significant pathological finding in the gross observations. Histological evaluations of disodium EDTA (60 mg/kg) group revealed an inflammatory lesions in the kidneys. However, in the recovery group did not show such lesions suggesting that the inflammatory lesions were transient and resolved upon discontinuation of treatment. It was concluded that repeated administration of disodium EDTA does not cause any observable toxicity.
