FORMULATION AND DEVELOPMENT OF KETOPROFEN NANOEMULSION AS A CARRIER FOR TOPICAL DELIVERY

ABSTRACT
The aim of the present investigation was todevelop and evaluate Ketoprofen loaded Nanoemulsion gel for treatment of Rheumatoid arthritis and osteo arthritis. Pseudo ternary phase diagrams were constructed for different Nanoemulsion formulations composed of Captex 200 as oil, Tween 20 as surfactant and PEG-400 as co-surfactant. Composition of Nanoemulsion system was optimized using 32 factorial design including the concentration of oil, concentration Surfactant and co-surfactant (Smix) and the % transmittance, viscosity and % drug release (3 hrs) as a dependent variable The physicochemical properties of the optimized microemulsion wer investigated. nanoemulsion was incorporated into 1% carbopol 934P gel and theex vivo drug permeation of gel was also investigated.
Theresults showed that the optimized nanoemulsion formulation was composed of Captex 200 oil (5.83%, w/w), Cremophor Tween 20 (53.83%, w/w). The mean globule diameter was 45.72 nm. The conclusion was that the release controllingability of nanoemulsion containing gel formulation was significantly improved in comparison to commercial gel. Skin irritation study was also performed and it shows no irritation on rat skin after 24 h of application.
