PENETRATION ENHANCERS: AN APPROACH TO INCREASE SKIN PERMEATION AND IT’S EFFECTS WITH EUTECTIC MIXTURE

ABSTRACT
Nowadays delivery of drugs through transdermal route are highly preffered due to their advantages over normal drug delivery routes. The most important parameter in delivery of drugs through skin is there penetration across skin crossing the stratum corneum barriers, there are various barriers which effects the permeation of drug into the skin,these may be either skin related factors or drug properties related factors. Hence to overcome these barriers the novel idea of penetration enhancers are genetrated. Penetration enhancers are substances which
are used to increase the permeationn of drug across the stratum corneum layer and also the various barriers. These may be physical, chemical or of natural origin.they are also known as absorption enhancers or permeation enhancers. An ideal penetration enhancer is one which have ideal properties to increase absorption.every penetration enhancer follow a systematic mechanism of action. They mainly act on site A (LIPID SITE), site B (AQUEOUS SITE), site C (LIPID DOMAIN). They should be adequately able to disrupt lipid layer, protein modification and partitioning promotion. It is also reported that if penetration enhancers are use in combination then they show synergistic effect and more rapid permeation of active drug through the skin.more over using penetration enhancers along with eutectic mixtures of drug makes it more efficient to deliver drug across the membrane.
