USED OPA-13 AND OPA-07 REPETITIVE SEQUENCE FOR DETECTION GENOMIC INSTABILITY IN EARLY STAGE COLON CANCER BY RAPD ANALYSIS

ABSTRACT
Most cancers of the colon and rectum display a phenomenon termed genomic instability. The technique that detects genomic alteration correlated with human tumor was microsatellites analysis. Opas primer was used in many cancer researches used two different 10- bases Opas primers to detect genomic instability in colon cancer. Method Genomic instability was detected by RAPD method By using two significant primers (OPE-07 and OPA-13). (12) patients after operation. the period March 2013 to April 2014 from the patients treated in Mosul Oncology and Nuclear Medicine Hospital, Jumhory Hospital/Mosul and, Azadi Teaching Hospital/Kirkuk. In all patients the diagnosis was confirmed histologically, which were classified as Duke's stage A, B, C and patients diagnosed as stage D. The mass and normal free tissues in addition to serum were taken from the same patients. The tissues were cut off, rinsed with normal saline, and kept frozen at (-20 Co) until analyzed. Results: The most important finding emerged from this study was serum utilization for early detection of colon cancer. Significant result was an insertion of bands at 500, 800 and 1000 bp amplified fragment observed in stage A tumor samples using primers OPE-07 while the amplified fragment was observed at 500, 600 and 700 bp in the same stage of colon cancer samples using primers OPA-13, these results indicate that there was a genomic instability detected in colon cancer by RAPD analysis Conclusions The most noteworthy finding of this study was the association between the level of genomic instability and colon cancers duck stages and the utilization of RAPD analysis.
