FORMULATION AND DEVELOPMENT OF BILAYER TABLETS OF CIMETIDINE AND PRAZIQUANTEL FOR TARGETED DELIVERY

ABSTRACT
In the present investigation bilayer tablets of cimetidine and
praziquantel were prepared. Cimetidine is anhistamine H2 antagonist
having synergistic effect with praziquantel and therefore it increases
the therapeutic efficiency of the praziquantel. Cimetidine mainly
locally acts on the stomach while praziquantel was targeted to the
colon for. So in the present study first layer of cimetidine float and
second layer of praziquantel was enteric coated. Individual
optimization was carried out for both the layer. For the floating layer
different swelling polymers like HPMC K4 M, HPMC K15 M and
HPMC E3 LV were used with different concentration (10, 20 and
30%). After the polymer optimization concentration of sodium
bicarbonate was optimized. Experimental design (CCD) was applied
for the floating layer optimization. For the enteric coating layer optimization was carried out
for different enteric coating polymers like Eudragit L100, Eudragit S100 and different
method of granulation like Intragranulation, Ladling. Final formulation for both the layer was
used for the final formulation of bilayer tablets. Different pre compression and post
compression parameters were used for the evaluation of the bilayer tablet. The bilayer tablets
were evaluated for in vitro drug release and both the layer were remained in the GIT for more
than 12 hrs. Final bilayer tablets were putted for stability study at the interval of 15, 21 and
30 days. The data of stability showed no any significant difference with the before data of the
tablets.
