Abstract

DESIGN AND EVALUATION OF MUCOADHESIVE BUCCAL TABLETS OF SUMATRIPTAN SUCCINATE

ABSTRACT

The present investigation was aimed at designing and evaluating bilayered buccal tablets of Sumatriptan Succinate by direct compression method using bioadhesive polymers like Hydroxy Propyl Methyl Cellulose (HPMC K4M), Carbopol 934p by combination of these polymers. Sumatriptan Succinate, which have well established efficacy in treating migraine. Sumatriptan Succinate was selected as a model drug for investigation, because of its very low oral bioavailability due to extensive first pass metabolism. All formulations were subjected to evaluation tests like thickness, weight variation, hardness, friability, disintegration, drug content, swelling studies, surface pH, and in-vitro drug release. The tablets were evaluated for in-vitro release in pH 6.8 phosphate buffer for 10hr in standard dissolution apparatus. In order to determine the model of release, the data was subjected to Korsmeyer and Peppas diffusion model. The optimized formula followed non-fickian release mechanism with zero order kinetics. The physicochemical interaction between the drug and polymers were investigated using Fourier transform infrared (FTIR) spectroscopy. The result of thickness, weight variation, hardness, drug content, was found to be within the limits. Optimized formulation containing HPMC K4M and Carbopol 934p showed 97% in-vitro release of drug in 10 hrs. The present study concludes that buccal delivery of Sumatriptan succinate tablets can be good way to bypass the first pass metabolism. It may render great bioavailability. In addition, these formulations reduce the need of frequent administration, dose dependent side effects and enhance patient compliance.
