A VALIDATED HPLC METHOD FOR THE ESTIATION OF GLIBENCLAMIDE NANOEMULSION FORMULATION IN RAT PLASMA: A PHARMACOKINETIC STUDIES

ABSTRACT
Glibenclamide is a second generation oral sulfonylurea antidiabetic agent widely used for the treatment of diabetes mellitus. The chromatographic separation of glibenclamide (GLBD) and internal standard (glipizide) were accomplished using 250 x 4.6 mm, C18 5 μm reverse phase analytical column. The mobile phase consisted of acetonitrile and 25mM monobasic potassium dihydrogen orthophosphate at 60:40 v/v ratios was run A rapid, sensitive, precise, accurate and specific HPLC assay for the determination of glibenclamide in rat plasma was developed and validated. The method was linear in the 10-400 ng/ml concentration range (r2 =0.999). The assay was used to estimation the pharmacokinetics of glibenclamide nanoemulsion gel formulation.
