Abstract

IN VITRO STUDIES ON INTERACTIONS BETWEEN CT-DNA AND NP- CHLOROPHENYL-2-FUROHYDROXAMIC ACID

ABSTRACT

The interaction of N-p-Chlorophenyl-2-Furohydroxamic Acid, (NCFHA) with calf thymus DNA (Ct-DNA) has been studied using absorption and emission spectroscopy, viscosity and docking. The results revealed decrease in absorption spectra, fluorescence quenching and enhancement of viscosity upon binding affinity to Ct-DNA. The binding constant Kb determined by absorption measurements for NCFHA- DNA complex was found to be 2.38 x 104 M-1 and Stern Volmer constant of complex is obtained as 8.94 x 102 M-1. The increase in viscosity and melting temperature (Tm) of DNA shows its interaction with the molecule. Docking was done using rigid modes of N-CFHA through Hex software package. The nature of binding and binding strength between N-CFHA and DNA is obtained from docking. Both experimental results and docking studies indicate the binding nature of N-CFHA bind with DNA.
