GENETIC POLYMORPHISM OF (CYP11B2) GENE IN HIGH BLOOD PRESSURE IRAQI PATIENTS

ABSTRACT
The present study was carried out in the Genetic Engineering and Biotechnology Institute – University of Baghdad between November 2014 - April 2015. 50 samples were collected from patients with high blood pressure in addition to 50 sample (control) of Yarmouk Teaching Hospital in Baghdad.Their ages different ranged from 20-60 years, to detect polymorphism (- 344 T / C) at the promoter of the gene region in CYP11B2 gene responsible for high blood pressure in Iraqi patients. Genomic DNA was extracted by using Bioneer DNA extraction kit from the whole blood. Conventional PCR and PCR-RFLP were used to detect the – 344 T/C in the CYP11B2 gene variant by using specific
primers and HaeIII restriction enzymes. The PCR fragment 538 bp if thymine T is at position – 344 the enzyme HaeIII is cut into three fragments (274, 138 and 126 bp ) , whereas fragments with cytosine C at the same position had the restriction site at (203, 138, 126 and 71 bp ). Results of the study showed that the incidence of high blood pressure disease in the age group 20-60 years was higher in males than in females, reaching (80% and 20%) respectively. The study showed that smoking, stress and weight factors all have a clear and serious impact on the proportion of people with high blood pressure. The results also showed that two mutations in the CYP11B2 gene cause high blood pressure. Results of statistical analysis mutation (-344 T / C) at the promoter of the gene region of the CYP11B2 gene between the healthy and the patients showed that the homozygous normal T / T is (38%) vs (22%) with a level of a (p˂ 0.01) significance while the heterozygous mutant T / C was (56%) vs (28%) which was of a (p˂ 0.01) significance. As for the homozygous mutant C / C the result was (34%) vs (22%) with a significance level of (p˂ 0.05) These results indicate that mutation may constitute a risk of developing high blood pressure.
