STUDY ON THE INCLUSION COMPLEX FORMATION OF DESIPRAMINE WITH ?-CYCLODEXTRIN AND ITS PARMACEUTICAL APPLICATION

ABSTRACT
The inclusion complex formation between desipramine (DES) and β–cyclodextrin (β–CD) was investigated by FT-IR and UV spectroscopy. The obtained results showed that DES reacts with β–CD to form a 1: 1 host – guest complex with an apparent formation constants of 8.92 x 103 L mol-1. Influence of pH and β-CD concentration on inclusion DES by β–CD was studied. Inclusion complex formation was also confirmed by scanning electron microscopy SEM. Based on enhancement of absorbance in presence of β-CD, a sensitive UV spectrophotometric method for determination of DES was developed. A linear relationship between absorbance and DES concentration in presence and absence of β-CD was obtained in the range of 3-55 μg mL -1 and 6-65 μg mL -1, with the correlation coefficient of 0.9996 and
0.9994, respectively. The proposed method was successfully applied to the determination of desipramine in commercial pharmaceutical preparations and serum. The results were satisfactory in comparison with the official and reported high performance liquid chromatography methods.
