EXTRACTION OF CAPSAICIN FROM Capsicum frutescens.L AND ITS ESTIMATION BY RP-HPLC METHOD

ABSTRACT
A new simple, precise, rapid, specific and accurate Reverse Phase High Performance Liquid Chromatography method was developed for Estimation of Capsaicin in Capsicum frutescens.L fruit extract. Extraction of chilies was performed by using Soxhlet apparatus, methanol as solvent. Chromatographic separation was performed using Hypersil BDS C8, 4.6x250mm, 5μm, Column with mobile phase comprising of Phosphate buffer pH-6.8, Methanol, ACN in the ratio of 30:60:10V/V/V, at a flow rate of 1.0ml/min. The detection was carried out at 281 nm. The retention times of Capsaicin were found to be 3.505min with a run time of 5min, theoretical plate counts for Capsaicin were 3164. As per ICH guidelines the method was validated for linearity, accuracy, precision, and robustness. The linearity was found in the concentration range of 50-150μg/ml. The correlation coefficients for Capsaicin were 0.998. This demonstrates that the
developed method is simple, precise, rapid, selective, accurate and reproducible for extraction of capsaicin from Capsicum frutescens.L and its estimation by RP-HPLC.
