FORMULATION AND EVALUATION OF BUCCAL MUCOADHESIVE TABLETS OF GLIPIZIDE

ABSTRACT
The present research work is aimed to prepare a buccal tablet for a low bioavailable anti diabetic drug glipizide. Preparing a buccal tablet not only improve the bioavailability but also aver comes the first pass effect. Bioadhesive polymers such as carbopol, chitosan, guargum and HPMC K15M have been employed in the present study. Suitability of the drug for the buccal delivery system has been studied and found that glipizide is suitable candidate and further studies have been carried out. Various formulations have been developed and evaluated for precompression and post compression parameters which revealed good flow properties of the blend and physical attributes of the prepared tablets were found to be practically within control limits. From the drug release studies it was found that formulation F3 containing carbopol has good drug release when compared to other formulations.
With respect to all the evaluation parameter formulation F3 was considered as the optimized one and is subjected to specific evaluation tests like swelling index, ex-vivo residence time, Moisture absorption, surface pH, Bioadhesion strength and found to be acceptable. The drug release was by zero order kinetics and drug release follows non-Fickian diffusion mechanism.
