Abstract

DESIGN AND EVALUATION OF GASTRORETENTIVE FLOATING DRUG DELIVERY SYSTEM OF ITOPRIDE HYDROCHLORIDE

ABSTRACT

Drugs that have narrow absorption window in the gastrointestinal tract will have poor absorption. For these drugs, gastroretentive drug delivery systems offer the advantage in prolonging the gastric emptying time. Itopride hydrochloride is an antacid drug; it has low elimination half life 4-5 hrs. The floating tablets of Itopride hydrochloride were prepared to increase the gastric retention and to improve the bioavailability of the drug. The floating tablets were formulated using HPMC K15M and Xanthan Gum as the release retardant polymers and sodium bicarbonate as the gas generating agent to reduce the floating lag time by using direct compression method. The formulated tablets were evaluated for pre-compression and post compression studies. The floating tablets extended the drug release up to 8 hrs. The FTIR study indicated the lack of drug-polymer interaction. The optimized formulation F2 containing drug HPMC K15M at 120 mg and Xanthan Gum 70 mg showed very good result and extended the release up to 12 hrs. The drug release from the optimized formulation followed Zero order kinetics (R2=0.993). The similarity factor calculated for batch F2 was 99.76%.
