Abstract

BIOASSAY-GUIDED FRACTIONAL ISOLATION AND IDENTIFICATION OF ANTI-MRSA COMPOUNDS BY GC-MS FROM TRIDAX PROCUMBENS L

Dr. Vilas A. Kamble* and Amit H. Moon

ABSTRACT

Introduction: The plant Tridax procumbens is well known for its pharmacological properties and ethano-botanical usages. Objective: The present study deals with the isolation and identification of anti- MRSA compounds as active secondary metabolites from the root part of Tridax procumbens L. Method: Antibacterial activity was assayed using agar well diffusion method. The metabolites profiling of bioactive extract was done by pre-coated silica gel TLC plates grade F254 & anti-MRSA compounds were located by TLC bio-autography (agar-overlay) assay. The characterization of the anti-MRSA compounds was carried out using the GC-MS analyser. Result: The methanolic root extract exhibited inhibition zone ranges from 17-23 mm against the clinical Methicillin Resistant S. aureus (MRSA) isolates originated from different hospitalised patients. TLC bioautography study revealed presence of two anti-MRSA compounds. These compounds were AM1 (Dasycarpidan-1-methanol, acetate ester) and AM2 (Milbemycin B, 6, 28-anhydro-15-chloro-25-isopropyl-13-dehydro-5-O-demethyl- 4-methyl) according to the mass spectra obtained from the GC-MS analysis and NIST data bank. Conclusion: The study concludes the plant had great potential for the growth inhibition of methicillin-resistant S. aureus and serves as potential source of anti-MRSA agents.

Keywords: Tridax procumbens L, TLC Bio-autography, GC-MS analysis, Solvent extracts, Secondary metabolites and anti-MRSA compounds.
