ABSTRACT

Shirishadi Compound is an Ayurvedic polyherbal preparation used in Indian traditional medicine for the treatment of Asthma & allergic disorders. The pharmacodynamic properties of ethanolic extract of Shirishadi compound was evaluated on experimental model for its antihistaminic property against H1 receptor and mast cell stabilizing property. Significant increase in preconvulsion time was observed on pretreatment with Shirishadi compound when the guinea pigs were exposed to histamine. Demonstration of graded dose response of histamine and its competitive antagonism on isolated perfused guinea pig ileum shows that it has specific antagonism. The increasing dose of Shirishadi showed the Tachyphylaxis and receptor protection response. The present study revealed that the ethanolic extract of Shirishadi compound has significant antihistaminic (H1 receptor antagonism) activity. Polyherbal formulation (500μg and 1000μg/ml) showed dose dependent significant reduction in mast cell degranulation (P
