Abstract

FORMULATION AND EVALUATION OF PIOGLITAZONE BUCCAL TABLETS

ABSTRACT

Aim of present study was to study and characterized mucoadhesive buccal tablets of pioglitazone hydrochloride using different mucoadhesive polymers such as chitosan,carbopol, HPMCK15M in combination. Twelve formulations were prepared with different concentrations of polymers individual and combination of two polymers in each formulation. Formulation F1 to F3 composed of individual polymers. F4 to F6 were composed of HPMCK15M, Carbopol as polymers. F7 to F9 formulations were composed of chitosan, carbopol as polymers. Where as formulation F10to F12 were composed of chitosan and HPMCK15M in the ratio of 1:2, 2:1, 1.5:1.5. The prepared tablets were evaluated for physicochemical parameters such as hardness, thickness uniformity, weight variation, surface pH, Ex-vivo residence time and moisture absorption studies. The prepared tablets were also evaluated for bioadhesive strength and in-vitro drug release. In-vitro bioadhesive strength and invitro release studies showed that formulation F9 containing 1.5:1.5 ratio of polymers combination and drug showed optimum drug release (98.72), and optimum bioadhesive strength. FTIR results showed no evidence of interaction between polymer and drug.
