ABSTRACT BACK GROUND AND THE PURPOSE OF STUDY: The objective of this study was to develop a controlled release matrix tablets of cefixime trihydrate(CFX) to deliver the drug at a constant rate. This is achieved by developing the formulations in the form of floating gastroretentive drug delivery system (FGDDS), which after Oral administration are designed to prolong the gastric residence time, thereby enhancing the absorption. METHODS: Cefixime trihydrate floating matrix formulations were prepared using two different polymers, hydroxypropyl methylcellulose (HPMC ) and polyethylene oxide (PEO). The matrix tablets were prepared by dry blending at selected ratios of polymer and other ingredients using direct compression method. RESULTS: Various parameters like invitro dissolution, matrix hydration, floating lag time, floating time and erosion studies were carried out. The kinetics of drug release, swelling and erosion of prepared matrix tablets were studied. Matrix tablets prepared using HPMC showed a greater degree of hydration and swelling when compared to formulation prepared by PEO. Drug release kinetics was evaluated using the Korsmeyer–Peppas model and found to be governed by non Fickian diffusion. CONCLUSION: Formulations prepared by blending drug, HPMC, PEO, gas generating agent and diluents followed by direct compression showed satisfactory results with respect to floating lag time, total floating duration, swelling ability and sustained drug release characteristics.
