ADVANCEMENTS AND CHALLENGES IN BUCCAL DRUG DELIVERY: A COMPREHENSIVE REVIEW

ABSTRACT
Buccal drug delivery systems offer a possible solution for conventional routes of administration, overcoming first-pass effect and affording quick absorption across the oral mucosa. It is especially useful for pediatric and geriatric patients since it provides ease to use, improving compliance. Prolonging drug retention at the application site is highly dependent on mucoadhesive polymers, and different formulation techniques such as hot-melt extrusion or solvent casting are used to improve functionality. While there are challenges of the low potency drugs and significant production cost, all those ongoing studies are focused to discover new formulations or improve already developed system. Here, in this review we highlights on anatomy of oral mucosa, drug permeation mechanisms which ultimately improve the rate and extent of biologic availability tests methods along with new advances for buccal mucoadhesive once-a day extended time delivery charting its clinical significance and future tendency.
