VORICONAZOLE A POWERFUL ANTIFUNGAL AGENT

ABSTRACT
Voriconazole which is chemically (2R, 3S)-2-(2,4-difluorophenyl)-3-(5- fluoro pyrimi din-4-yl) -1- (1,2,4-triazol-1-yl) butan- 2-ol.Voriconazole is a triazole antifungal agent. The primary mode ofaction of voriconazole is the inhibition of fungal cytochrome P-450-mediated 14 alpha-lanosterol demethylation, an essential step in fungalergosterol biosynthesis. The accumulation of 14 alpha-methyl sterolscorrelates with the subsequent loss of ergosterol in the fungal cell walland may be responsible for the antifungal activity of voriconazole.Voriconazole has been shown to be more selective for fungalcytochrome P-450 enzymes than for various mammalian cytochromeP-450 enzyme systems. Invasive aspergillosis and serious fungalinfections due to Fusarium spp. and Scedosporium apiospermum.Literature survey reveals that Voriconazole is estimated individuallyby UV spectrometry, RP-HPLC, HPLC-UV, RP-UPLC, LC/MS/MS,HPTLC and stability indicating UPLC.
