FORMULATION AND EVALUATION OF MUCOADHESIVE TABLETS OF DILTIAZEM HYDROCHLORIDE

ABSTRACT
Mucoadhesive drug delivery systems are among the popular drug delivery systems designed to combat bioavailability problems of drug undergoing extensive first pass metabolism when administered orally. Diltiazem was selected as a model drug for investigation because of its suitable properties like half-life of 4.5 hrs optimum partition coefficient and molecular weight make it suitable for administration by buccal route. Diltiazem hydrochloride Mucoadhesive buccal tablets were prepared by using carbopol as a primary polymer because of its excellent mucoadhesive property and secondary polymers like HPMC, SCMC, sodium alginate and guar-gum were used. Formulations were developed with varying concentrations of polymers. The effect of secondary polymer loading on drug release was studied. Theformulations were tested for in-vitro drug release and in-vitro swelling studies. Formulation FD4 showed maximum release of 83.23% in 8 hrs. Carbopol shows satisfactory bioadhesive properties Formulation FD4 using HPMC and Carbopol polymer (1:2) ratio showed significant bioadhesive properties with an optimum release profile and could be useful for buccal administration of diltiazem. The all synthetic polymers shows good bioadhesive properties as compared to natural polymer.
