Abstract

FORMULATION AND EVALUATION OF BILAYERED FLOATING TABLETS CONTAINING METFORMIN SR AND PIOGLITAZONE IR

ABSTRACT

The present work focuses on Formulation and Evaluation of bilayered floating tablets containing Metformin for sustained release and Pioglitazone for immediate release. Sustained release layer was prepared by wet granulation method using HPMC K4M, HPMC E50 as release retarding polymers. Immediate release layer was prepared by direct compression method using sodium starch glycolate as super disintegrant. Bilayered floating tablets were evaluated for Hardness, Friability, Weight variation, Drug content,Floating properties, Water absorption ratio, Wetting time, In- vitro release studiesand the values were found to be satisfactory. All the tablets showed goodfloating lag time(68-130sec).In vitro release studies were carried out in 0.1N HCl using USP apparatus ІІ. The release kinetics of Metformin was evaluated using the regression coefficient analysis. Among all the formulations FF4 was the best formulation showed slow sustained release(98.73%) upto 12hrs. Non-Fickian diffusion was the drug release mechanism and it is fallowing zero orderkinetics. FT-IR studies suggesting that there is compatibility between the drug and excipients.
