Aditya Santosh Ambi*, Swati Rawat, Pravin Wakte and Sachin Bhusari

ABSTRACT

In situ gel drug delivery systems represent a promising approach to overcome the limitations of conventional dosage forms by providing localized and sustained drug release at the site of administration. This review explores the diverse gelation mechanisms, various approaches, evaluation and recent advancements in in situ gel systems across various therapeutic areas. Highlighting their critical roles in optimizing therapeutic efficacy and patient compliance. Furthermore, the integration of stimuli-responsive materials and nanotechnology in designing smart in situ gels for targeted drug delivery is examined, emphasizing their potential to enhance therapeutic outcomes while minimizing systemic side effects. Challenges including gelation Kinetics, biodegradation, manufacturing scalability, and tissue Adhesion are also addressed, underscoring the need for continuedinterdisciplinary research to advance these systems from bench to bedside. Ultimately, this review aims to provide a comprehensive overview of current trends, challenges, and future perspectives in in situ gel drug delivery systems, advocating for their potential to revolutionize personalized medicine and improve patient outcomes.

Keywords: In situ gelling systems, drug delivery, polymers, thermo-sensitive systems, ion-sensitive systems, pH-sensitive systems, muco-adhesion, non-parenteral administration routes.
