Abstract

FORMULATION AND EVALUATION OF FAST DISSOLVING BUCAL FILMS OF PHENYLPROPANOLAMINE HCL

Pramod M. V.*, Suresh V. Kullkarni and K. Manjunath

ABSTRACT

Phenylpropanolamine HCI is commonly found in appetite suppressants, used to treat cough, cold, haemorrhagic stroke. Phenylpropanolamine hydrochloride is widely used as a decongestant and an anorectic agent. As of phenylpropanolamine HCl undergoes first pass metabolism, development of fast dissolving buccal films of phenylpropanolamine HCl release the drug in the buccal cavity and absorb through buccal region. Hence, first pass metabolism of the drug could be avoided by developing into fast dissolving buccal film of phenylpropanolamine HCl. This is generally achieved by drug to be administered directly into the blood circulation through the buccal mucosa. The drug administered in the buccal route can be metabolized more through GIT hence buccal film gives fast onset of action. The objective of this research work was to improve the bioavailability of the drug and fasten the onset of action. The buccal bioavailability of phenylpropanolamine HCl is only 38% for this reason drug can be administrated into buccal mucosa using mucoadhesive film. It can be made fast drug release and directly enter into blood circulation through buccal mucosa and increase the drug bioavailability.

Keywords: Phenylpropanolamine HCl, HPMC E 15, PVP K-30, PVA, Vanillin, citric acid, PEG 400, Solvent casting method.
