ABSTRACT
Oral drug administration is the most widely used means of drug delivery in the pharmacy field. It has dominated other drug delivery systems for advantages like ease of administration, flexibility in formulation, patient compliance, etc. However, oral administration has demerits too. Oral drugs don’t get sufficient residence time in stomach, they are non site-specific, less soluble in alkaline pH and show low bioavailability. To overcome such problems faced by oral administration, gastro retentive drug delivery system (GRDDS) were developed. GRDDS is a novel approach to retain the dosage form in the stomach for an extended period of time and release the drug slowly. Prolonged gastric retension addresses many challenges associated with conventional oral drug delivery that are discussed in this article. This article provides concise information about gastro retentive drug delivery systems with special focus on approaches, needs, factors controlling gastric retention, advantages, disadvantages, applications and future potential for GRDDS.
