FORMULATION AND EVALUATION OF EXTENDED RELEASE TABLET OF TOLMETIN BY MELT GRANULATION TECHNIQUE

ABSTRACT
In present study, extended release tablet of Tolmetin has been prepared by melt granulation method using carnauba wax and bees wax in different concentrations. Pre-compression parameters of powdered blend and extended release tablets were evaluated and found passes as per specifications. The result of FTIR study shows that there was no interaction between drug & polymers. In vitro drug release studies in 0.1 N HCl (First 2 h) and 6.8 pH phosphate buffer show drug release from range 92.3±0.23% to100.2±1.36% all formulation, Batch T6 indicated a higher release rate of 100.2% in 24 hours containing bees wax and follows zero order kinetics as correlation coefficient (r2) values are higher than that of first-order release kinetics. The release exponent value (n) obtained was 0.82105. The T6 formulation exhibited anomalous (non- fickian) diffusion mechanism. Kinetic studies shows that the amount of drug from the matrix system was by both diffusion and erosion. The optimized formulation (T6) did not show any significant change in appearance, hardness, friability, thickness and % drug content and showed stable retaining their original properties which indicates no alteration after storage.
