AUGMENTATION OF TNF-?, COX-2, CASPASE-3, CD95 EXPRESSION IN MICE LIVER CAUSED BY AZORUBINE ADMINISTRATION AND THE IMMUNOMODULATORY EFFECT OF VITAMIN E

ABSTRACT
The present study aimed to evaluate the mechanism of action for
vitamin E against hepatotoxicity by food additive azorubine in male
young mice. Mice divide into 4 groups: group I was the control; group II
received vitamin E (100mg/kg); group III received azorubine
(250mg/kg) and group IV received vitamin E+azorubine for 28 days.
Hepatotoxicity occurred as a result of increased hepatic markers,
changes in antioxidant activities, increased of TNF-α, COX, caspase-3,
CD95 and decreased of Bcl-2 concentrations. Hepatic tissue showed
degeneration and pyknosis due to the administration of azorubine.
Injection of vitamin E caused significant restoration in the levels of
liver functions, antioxidants status, histological architecture in liver \
and improvement in the immunological parameters (TNF-α, Cox-2, caspase-3, CD95 and
Bcl-2). The preventive role of vitamin E against azorubine induced toxicity attributed to its
powerful antioxidant role, free radical scavenging properties and its immunomodulatory
effect.
