FORMULATION AND EVALUATION OF FAST DISSOLVING TABLETS OF ANTIEMETIC DRUG METOCLOPRAMIDE

ABSTRACT
Metoclopramide hydrochloride(MH) is chemically 4-amino-5-chloro-
N[2-(dimethylamino)-ethyl]-2-methoxybenzamide monohydrochloride
monohydrate. It is a potent dopamine receptor antagonist. It is a potent
antiemetic and is effective in the treatment of nausea and vomiting
associated with cancer chemotherapy, pregnancy, migraine etc. It is
also used for the treatment of diabetic gastric stasis and gastrooesophageal
reflux disease. The fast dissolving tablet was prepared by
using different superdisintegrant with different ratio. FT-IR study
revealed no interaction between the drug and excipients. Tablets were
characterized for hardness, friability, weight variation, wetting time, disintegration time, drug
content, dissolution and dispersion time. Among the nine formulations, F6 was selected as the
best formulation as its wetting time was 39 seconds, disintegration time was 12 seconds,
dispersion time was 49 seconds and %CDR after 6 minutes was 99.46%. F6 was found to be
stable at 40 °C ± 2 °C and 75 ± 5 % RH which was confirmed by FT-IR study.
