EFFICIENT POLYHYDROXYBUTYRATE PRODUCTION FROM SOIL BACTERIAL SP, USING LOW COST CARBON SOURCE FOR PRODUCTION OF ECO FRIENDLY BIOPLASTICS

ABSTRACT
In this present study soil bacteria were isolated. The isolates were subjected to gram staining and biochemical analysis for identification. The screening of soil bacteria for PHB production was followed by Sudan black dye method & and light microscope method, that showed two isolates were showed positive result. That were black colonies on morphology & black colour cell appearance under light microscope. The soil bacteria B1 for PHB production showed that yield 0.18g/100ml for 48hrs fermentation period. Then the optimization of Polyhydroxybutyrate production was studied at various incubation period, pH, temperature, carbon & nitrogen source. This revealed the PHB production was better at 48hrs incubation period, pH at7, temperature 37oC, carbon source was sucrose & nitrogen source was
ammonium chloride. The FT-IR spectrum showed that functional group of PHB.
