ABSTRACT
Smart nanocarrier-based drug delivery systems, sometimes referred to as Smart Drug Delivery Systems (SDDSs), were developed as a result of the nonspecific dispersion and unpredictable release of pharmaceuticals in conventional drug delivery systems (CDDSs). To lessen the negative effects associated with CDDSs, SDDSs can administer medications to the target areas with reduced dosage frequency and in a spatially controlled way. Cancer, the second-leading cause of death worldwide, is frequently treated with chemotherapy. An intense interest in the SDDSs as an alternative to chemotherapy resulted from site-specific drug delivery. The focus of
SDDSs is on smart nanocarriers, which are nanoparticles used to transport medications. Smart nanocarriers, targeting mechanisms, and stimulus methods make up a smart medication delivery system. The current development is highlighted in this review. Several smart nanocarriers, including liposomes, micelles, dendrimers, meso-porous silica nanoparticles, gold nanoparticles, super paramagnetic iron-oxide nanoparticles, carbon nanotubes, and quantum dots, have recently undergone the creation of SDDSs. The architecture, classification, synthesis, and level of intelligence of the nanocarriers are detailed. There are significant worries about the toxicity of smart nanocarriers despite the fact that SDDSs have a lot of advantages over chemotherapy. As a result, a thorough investigation into the toxicity and biocompatibility of the nanocarriers has been described. Finally, the difficulties and potential areas for future research in the field of SDDSs are also discussed. This review is anticipated to be extremely helpful for both individuals who are just beginning their studies in smart nanocarrier-based medication delivery as well as those who have been looking for fresh research avenues in this area.
