Abstract

DEVELOPMENT AND VALIDATION OF SIMULTANEOUS DERIVATIVE SPECTROPHOTOMETRIC METHOD FOR DETERMINATION OF SILDENAFIL CITRATE AND TRAMADOL HYDROCHLORIDE IN SYNTHETIC MIXTURE

ABSTRACT

Simple, accurate, precise, economical and reproducible analytical method has been developed for the simultaneous estimation of Sildenafil citrate and Tramadol hydrochloride in synthetic mixture form by UV spectrophotometric first order derivative method. The stock solutions were prepared in methanol. In the first order derivative method, the wavelengths at which SIL and TRA were analyzed were 267.7 nm and 309.5 nm respectively. At 249.6 nm SIL has absorbance while TRA shows zero absorbance. Similarly, at 297.2 nm SIL shows absorbance while TRA has zero absorbance. Thus, both the drugs do not interfere in the quantitation of one another. Rom appropriate dilutions of the working standard stock solution, 10 µg/mL and 20 µg/mL concentration of Sildenafil citrate (SIL) and Tramadol hydrochloride (TRA) were separately prepared and scanned in the UV range 200–400 nm. The linearity was obtained in the concentration range for SIL 5-25 µg/mL and for TRA 20-100 µg/mL. The percentage RSD for precision and accuracy of method was found to be less than 2 %. Therefore, proposed methods can be successfully used for routine analysis of Sildenafil citrate and Tramadol hydrochloride in bulk as well as synthetic mixture.
