STUDY OF PREFORMULATION CHARACTERIZATION AND UV METHOD DEVELOPMENT OF ELLAGIC ACID

ABSTRACT
Ellagic acid (EA) which is obtained from various fruits and vegetables chemically is a polyphenolic compound which has been reported to show wide variety of pharmacological activities. The main objective of the present work is the determination of preformulation characteristics of ellagic acid (EA) including development of Ultra-violet (UV) spectrophotometric method of estimation. The preformulation studies of EA are performed for the analysis of solubility, melting point, partition coefficient, Fourier - transform infrared spectra (FTIR), X – ray powder diffraction (XRD) and differential scanning colorimetry (DSC) studies. The method of estimation was conducted by Absorption
maxima method. EA is poorly soluble in water and freely soluble in PEG 400 and dichloromethane. The partition co efficient, log P of ellagic acid is 2.21 which indicates good permeation across the lipid bilayer membrane. FTIR revealed O-H stretching at 3558.51 cm-1, phenyl C-O stretching at 1117.90 cm-1 and ortho disubstituted aromatic sp2 C-H bending at 760.66 cm-1. XRD indicated highly crystalline nature. DSC showed melting peak at 381.07 o C. The proposed UV spectrophotometric method of estimation of ellagic acid in PEG 400 was simple, convenient and accurate. Preformulation studies of the present work can be used for the design of ideal dosage forms of EA.
